Bacterial Disinfectant Efficacy Using Flow Cytometry

ثبت نشده
چکیده

Flow cytometry is a rapid method to quantitate live and dead bacteria in a sample.1-4 It can be used to measure the efficacy of antimicrobial compounds by exposing organisms to the disinfectant and subsequently staining with permeant and impermeant DNA dyes. Test organism viability, measured using a 30-minute flow cytometry assay, showed comparable sensitivity with a standard 48-hour plate count assay. Flow cytometry provides a rapid and quantitative method to determine the efficacy of disinfectants.

Comparison between Flow Cytometry and Traditional Culture Methods for Efficacy Assessment of Six Disinfectant Agents against Nosocomial Bacterial Species

The present study was undertaken to compare the use of flow cytometry (FCM) and traditional culture methods for efficacy assessment of six disinfectants used in Quebec hospitals including: two quaternary ammonium-based, two activated hydrogen peroxide-based, one phenol-based, and one sodium hypochlorite-based. Four nosocomial bacterial species, Escherichia coli, Staphylococcus aureus, Pseudomon...

Near Real-Time Flow Cytometry Monitoring of Bacterial and Viral Removal Efficiencies during Water Reclamation Processes

Wastewater reuse has become an important part of the urban water supply portfolio in water stressed regions. Effective wastewater treatment processes are critical to protect public health during water reuse practices. However, the microbial removal efficiencies in wastewater reclamation plants are not routinely monitored due to the lack of a simple quantification method. This study applied a ne...

Flow cytometry analysis of the activity of disinfecting agents tested with Staphylococcus aureus and Escherichia coli.

The efficacy of five disinfecting agents was tested by a new microbiological method, i.e. the flow cytometry. The method is based on kinetic measurements of the effects of a disinfecting agent on the percent of live/dead cells detected with propidium iodide. E. coli ATCC52922 strain and S. aureus ATCC 29213 strain were used in the experiment. From the measurements the killing time 50 (KT50) was...

Effect of sodium hypochlorite on typical biofilms formed in drinking water distribution systems.

Human health and biological safety problems resulting from urban drinking water pipe network biofilms pollution have attracted wide concern. Despite the inclusion of residual chlorine in drinking water distribution systems supplies, the bacterium is a recalcitrant human pathogen capable of forming biofilms on pipe walls and causing health risks. Typical drinking water bacterial biofilms and the...

The efficacy of a foaming iodine-based pre-milking teat disinfectant

Pre-milking teat disinfection is practised in several countries to prevent environment-related mastitis. This study was designed to prove the antimicrobial efficacy of a foaming, iodine-based teat disinfectant with five different concentrations (250; 500; 1,000; 2,000 and 3,000 ppm) against a negative control. For this purpose the split-udder design was used and within an udder two teats were d...
